Investigation of Intraspecific Variations in Campanula takesimana by RAPD DNA Markers

Abstract
RAPD markers were used to identify the variations of 13 lines of Campnula takesimarca and C. punctata. Of forty arbitrary primers tested, ten primers revealed a total of ninety different DNA fragments of 25 monomorphic bands and 65 polymorphic bands. The sizes of amplified DNA fragments were in the range of 400 to 2700bp. As the results of cluster analysis based on the DNA polymorphism, they were divided into five groups at the similarity coefficient value of 0.68. C. punctata was distinctly different from the intraspecific groups of C. takesimana. On the other hand, there was high relationship among intraspecific groups of C. takesimana. A few lines which showed distinctly different characteristics were classified by RAPDs, but genetic relationships based on RAPDs were not clear in accord with external characteristics.
